Enzo Life Sciences的ROS-ID® Superoxide detection kit可利用熒光顯微鏡和/或流式細(xì)胞儀直接實(shí)時(shí)監(jiān)測(cè)活細(xì)胞中超氧化物的產(chǎn)生。該試劑盒的一個(gè)主要成分,超氧化物檢測(cè)試劑(橙色),是一種細(xì)胞滲透性探針,可與超氧化物發(fā)生特異性反應(yīng),產(chǎn)生橙色熒光產(chǎn)物。熒光探針對(duì)活性過氧化物、羥基、過氧亞硝酸鹽、氯或溴相對(duì)不敏感,因此該試劑盒不能檢測(cè)這些分析物。染色后,可使用配備有標(biāo)準(zhǔn)橙色(如550/620 nm)濾光片的寬場(chǎng)顯微鏡進(jìn)行觀察,或使用配備有藍(lán)色(488 nm)激光的流式細(xì)胞儀進(jìn)行分析。
超氧化物檢測(cè)試劑盒包含足夠的試劑,可使用活細(xì)胞(貼壁或懸?。┻M(jìn)行至少200次顯微鏡分析或50次流式細(xì)胞術(shù)分析。
