Enzo Life Sciences的CYTO-ID®綠色/紅色長(zhǎng)期細(xì)胞示蹤試劑盒采用專有的非共價(jià)細(xì)胞標(biāo)記技術(shù),將含有疏水性脂肪鏈的熒光染料穩(wěn)定地結(jié)合入細(xì)胞膜的脂質(zhì)雙分子層中。標(biāo)記緩沖液對(duì)哺乳動(dòng)物細(xì)胞是等滲的,不含去污劑或有機(jī)溶劑。染料對(duì)細(xì)胞沒(méi)有毒性,可在細(xì)胞中保留長(zhǎng)達(dá)96小時(shí),并在有絲分裂時(shí)傳遞給子細(xì)胞。由于該染料不對(duì)細(xì)胞內(nèi)的蛋白質(zhì)進(jìn)行共價(jià)修飾,與基于硫醇反應(yīng)的氯甲基或胺反應(yīng)的琥珀酰亞胺酯類熒光染料的分子探針相比,細(xì)胞正常的生理反應(yīng)得到更好的保留。
也可以使用CELLESTIAL®染料進(jìn)行雙重標(biāo)記。標(biāo)記的細(xì)胞可以通過(guò)共聚焦熒光顯微鏡等儀器進(jìn)行觀察。此外,染料標(biāo)記的和未標(biāo)記的細(xì)胞群可通過(guò)流式細(xì)胞儀進(jìn)行分析。在延長(zhǎng)96小時(shí)的培養(yǎng)后,沒(méi)有觀察到熒光轉(zhuǎn)移到相鄰細(xì)胞。這與Calcein AM和BCECF AM標(biāo)記形成鮮明對(duì)比,后者在生理溫度下只能在存活的細(xì)胞內(nèi)保留幾個(gè)小時(shí)。
